Generation of a Three-Dimensional in Vitro Ovarian Cancer Co-Culture Model for Drug Screening Assays

In vitro 3D culture models have emerged in the cancer field due to their ability recapitulate characteristics of vivo tumor. Herein, we described establishment and characterization multicellular spheroids using ovarian cells (SKOV-3) co-culture with mesenchymal (MUC-9) or fibroblasts (CCD27-Sk). We demonstrated that SKOV-3 were able form regular compact diameters ranging from 300 400 µm a roundness close 1.0 regardless type stromal cell used. an increase was not observed spheroid diameter nor there significant growth. What is more, co-cultures presented up regulation genes related tumorigenesis, angiogenesis metastases (MMP2, VEGFA, SNAI1, ZEB1 VIM) when compared 2D monoculture. As expected, both cultures (mono co-cultures) exhibited higher Paclitaxel chemoresistance condition. Although did observe differences resistance between mono co-cultures, gene expression results indicate presence better tumor microenvironment, being able, therefore, more accurately evaluate drug efficacy for therapy.
